Polymerase Chain Reaction (PCR)
PCR is a targeted molecular method that amplifies selected DNA sequences. Related methods, including reverse-transcription PCR and quantitative PCR, can analyse RNA targets or measure the amount of a specific target.
Common Applications
- Detecting selected genetic variants or fusion transcripts.
- Supporting diagnosis and monitoring in selected haematological conditions.
- Detecting specific infectious agents when a validated assay is available.
Samples and Assays
The correct specimen depends on the clinical question and assay. Blood, bone marrow, tissue, swabs, or other samples may be appropriate. Pre-analytical handling and target quality can affect the result.
Important Limitations
PCR usually examines a defined target. A negative result does not exclude changes or organisms outside the assay’s scope, and results should be interpreted with clinical and laboratory findings.
